I will be changing the GPU card in one of my boxes from a NVIDIA GTX-760 to a Radeon RX-580. Will I need to clear out my GPU work unit cache before the change?

Yes, your BOINC client will download a different app for Radeon (opencl_ati_101), so the best thing to do is to finish all your current Nvidia tasks (opencl_nvidia_101) and then install the new card
